FOXO4 itself is a transcription factor protein involved in cellular regulation. The experimental compound typically referred to in this context is FOXO4-DRI, a synthetic peptide designed to interact with this pathway.
FOXO4-DRI is classified as a "senolytic" agent, meaning it is designed to selectively induce the death of senescent cells. Cellular senescence is a state where cells stop dividing but remain metabolically active, often secreting inflammatory molecules. The accumulation of these cells is considered a hallmark of aging and contributes to various age-related diseases.
The proposed mechanism of action for FOXO4-DRI involves disrupting the interaction between the FOXO4 protein and the p53 tumor suppressor protein. This FOXO4-p53 complex is believed to function as a pro-survival signal that specifically prevents senescent cells from undergoing apoptosis (programmed cell death). By breaking this link, FOXO4-DRI is intended to selectively trigger apoptosis in senescent cells while leaving healthy, non-senescent cells largely unaffected. Preclinical research in animal models has suggested this clearance can ameliorate some age-related physiological declines.
